The clinical aspect is characterized by a solid tumefaction of the right buttock and the right lower limb. Ultrasound and MRI showed the presence of an ill-defined mass on the right sacroiliac joint with local infiltration of the ligaments, the muscle and the bone. The tumour was characterized by the presence of atypical spindle cell proliferation with a biphasic architecture and local osteolysis. Immunohistochemical study showed that tumour cells were positive for epithelial and muscle markers and negative for lymphoid and neural markers. Retinoic acid has been found to inhibit both the growth of human colorectal cancers in vitro and tumor growth in vivo. We have investigated the effect of all-trans-retinoic acid on the production of a plasminogen activator by HCT-8 human colorectal cancer cells. Incubation of HCT-8 cells for 24 hours with 40 microM all-trans-retinoic acid produced a time- and dose-dependent inhibition of urokinase production (IC50 = 25 microM). In contrast, another human colorectal cancer cell line, SW-480, was much less sensitive to retinoic acid. We found that growth of HCT-8 cells was inhibited by all-trans-retinoic acid, and this inhibition was correlated with a decrease in urokinase production. Retinoic acid-treated cells produced less urokinase and showed a decrease in the catalytic activity of urokinase as assayed with a synthetic substrate. The decrease in urokinase was not related to a decrease in the mRNA content of the urokinase gene.
